The Environmental Research and Innovation (ERIN) department, made up of 200 life science, environmental science and information technology researchers and engineers, provides the interdisciplinary knowledge, expertise and technologies to lead solutions including the major environmental challenges facing society, such as climate change mitigation, ecosystem resilience, sustainable energy systems, efficient use of renewable resources, and environmental pollution prevention and control.

The department relies on two cutting-edge platforms, the Biotechnologies and Environmental Analytics Platform and the Observatory for the Climate and the Environment, and the GreenTech Innovation Centre (GTIC): a one-stop-shop for the complete development of bio-based products and processes.

This internship project is focused on fungal genetics and molecular biology aiming at implementing different steps of the avant-garde genome sequence-independent methods for heterologous expression of intact biosynthetic gene clusters (BGCs). Genome sequence-independent methods for heterologous expression of entire BGCs have only been possible recently with the latest advances in molecular biology tools. These methods make use of expression libraries on sheared genomes obtained from environmental DNA or from pure microbial/cell cultures. Therefore, the intern will have the opportunity to develop their skills in modern molecular/synthetic biology techniques whilst working with fascinating filamentous fungi.

This internship is proposed to be developed in a 6-months project focusing on the heterologous expression of intact fungal secondary metabolite (BGCs) in Aspergillus nidulans.

Responsibilities

How will you contribute?

The intern will:

  • Establish reliable protocols for extraction and isolation of high-quality high-molecular weight DNAs from pure cultures and environmental samples
  • Establish an unbiased method for DNA fragmentation
  • Set up and optimise a cloning strategy using fungal artificial chromosomes (FACs)
  • If time allows, optimise FAC transformation of Aspergillus nidulans for heterologous production of secondary metabolites


At the end of the 6-months internship the student is expected to have acquired knowledge in microbial cell culture (fungi and bacteria) and molecular biology, as well as basic skills in fungal biology and genetics.

You hold a MSc or this is your last year of BSc in one of the following areas: microbiology, biotechnology, industrial biotechnology, biochemistry, molecular biology or related fields.

You also have:

  • Some knowledge of microbiology, biochemistry and molecular biology (highly desirable)
  • Good knowledge of English
  • Previous laboratory experience will be an advantage
